Tween-20 is a non-ionic detergent with a molecular weight of about 600 and a critical micelle concentration (CMC) of 0.06-0.07% in water at room temperature. Its working mechanism may in part be based on its action as a detergent, binding to the hydrophobic moieties of water insoluble compounds, rendering them hydrophilic. In addition, immuno-compounds may become incorporated into micelles when the Tween-20 concentration is higher than the CMC, for instance at 0.1 % in PBS at pH 7.4.

Aurion has developed Blocking Solutions which effectively block ‘sticky' surfaces by multi-point hydrophobic and charge-based interactions. The Blocking Solutions are specifically designed to meet the characteristics of each type of secondary conjugate. With adequate blocking and incubation media, background reactions are minimized whereas specific reactions are not hampered.

The 3KC533 Heterophilic Blocking Reagent 1, Purified from Scantibodies Laboratory, Inc. contains specific murine immunoglobulins that block the heterophilic interaction by active binding to the heterophilic antibodies, which are capable of cross-linking the capture and the detection antibodies used in the immunoassay, resulting in false positive readings. The attachment of HBR-1 to the heterophilic antibodies blocks this cross-linking, and eliminates the interference caused by the heterophilic antibodies in the humoral fluids. In addition to its active blocking, this product is also characteristic of its passive blockage of the heterophilic interaction as well. The HBR is a liquid reagent with a protein concentration of 20 ± 2 mg/ml. The immunoglobulins are dissolved in a phosphate buffer with a pH of 7.2-7.6. The immunoglobulins in this product are at a purity of greater than or equal to 95%.

Western Blot Blocking Solution is specifically designed for western blotting using fluorochrome conjugated antibodies. Pure nitrocellulose membrane is recommended for maximum performance. Other membranes, such as PVDF or nitrocellulose embedded in a support can be used, but may generate elevated backgrounds. Protein should be transferred from gel to membrane using standard protocols. Blocking buffer can be used for membrane blocking and to dilute both primary and secondary antibodies. AURION R-Gent SE-EM is a high efficiency silver enhancement reagent for electron microscopy. It intensifies the gold particles by homogeneous deposition of metallic silver on the particle surface. The resulting electron dense signal is easily detected and is compatible with heavy metal counterstaining. The reagent has extremely delayed auto-nucleation and can be used under standard laboratory light conditions. It also has low viscosity, which is especially advantageous for pre-embedding immunogold applications.

It has a concentration of 10.0 +/- 1.0 mg/ml.
