Biotium DIHYDROETHIDIUM (HYDROETHIDIUM)
Dihydroethidium (also called hydroethidium) is the chemically reduced form of the commonly used DNA dye ethidium bromide. The probe is useful to detect oxidative activities in viable cells, including respiratory burst in phagocytes. Dihydroethidium itself has blue fluorescence Ex/Em: 355/420nm) in cells, while the oxidized form ethidium has red fluorescence Ex/Em: 518/605 nm) upon DNA intercalation.Properties: Off-white to light brown solid soluble in DMF or DMSO; Store at -20°C under nitrogen or argon, and protect from light, especially when in solution; C21H21N3; MW: 315; [38483-26-0]

Biotium PAGE GelRed Nucleic Acid Gel Stain, 10,000X in Water
PAGE GelRed is a non-toxic, non-mutagenic red DNA gel stain specifically designed to stain DNA in polyacrylamide gels. PAGE GelRed can be imaged using a 254 nm UV transilluminator with an ethidium bromide filter. While PAGE GelRed also can be used to stain DNA in agarose gels, Biotium’s original GelRed nucleic acid gel stain is more sensitive for agarose gel applications. PAGE GelRed can be removed from DNA after agarose gel staining using commonly available gel extraction kits. PAGE GelRed is 6-8 times more sensitive for dsDNA over RNA. Properties: Liquid; Ex/Em= 308, 551/625 nm (with dsDNA); Store at room temperature and protect from light.

Biotium Hoechst 33258, 10mg/mL in H2O
Hoechst dyes are cell membrane-permeant, minor groove-binding blue fluorescent DNA stains. These dyes are widely used in cell cycle and apoptosis studies as nuclear counterstains. Hoechst 33528 is slightly more water soluble than Hoechst 33342. The dyes can be used to stain live or fixed cells in buffer or medium at 1 ug/mL, with no wash step required. Properties: Yellow solution; Store at 4°C, protect from light; Ex/Em= 352/461 nm (with DNA); C25H27Cl3N6O; MW: 533.88; [23491-45-4].

Biotium DMAO, 2mM in DMSO
DMAO is a membrane-permeable DNA dye, excellent for staining live bacteria. Stained cells can be detected by fluorescence microscopy or flow cytometry. DMAO preferentially binds to dsDNA with green fluorescence (Ex/Em, DNA-bound = 496/528 nm). The dye is suitable for both gram-positive and gram-negative strains. Unlike the SYBR dyes, DMAO is extremely stable and can be routinely handled and stored at room temperature without degradation. Properties: Liquid; Store at room temperature or below, protect from light; Ex/Em = 496/528 nm (with DNA).

Biotium GelGreen Nucleic Acid Gel Stain, 10, 000X in Water, 10 mL
GelGreen is a sensitive, stable and environmentally safe green fluorescent nucleic acid dye designed to stain either dsDNA, ssDNA or RNA in agarose gels. GelGreen is far more sensitive than SYBR Safe. Unlike SYBR dyes, which are known to be unstable, GelGreen is very stable, both hydrolytically and thermally. GelGreen is compatible with either a 254 nm UV transilluminator or a gel reader equipped with visible light excitation (such as blue LED light box, 488 nm laser-based gel scanner, or Dark Reader). GelGreen can be used for either post-electrophoresis gel staining or precast gel staining. Properties: Liquid; Store at room temperature and protect from light.

Biotium NucSpot 470, 1000X in DMSO
NucSpot 470 is a cell membrane-impermeant green fluorescent DNA stain. It is virtually non-fluorescent in the absence of DNA, but fluoresces bright green upon DNA binding. While other green nucleic acid stains like TOTO, TO-PRO, or SYTOX dyes stain both the nucleus and cytoplasm, NucSpot 470 specifically stains the nucleus of fixed and permeabilized cells. It also can be used to selectively stain dead cells in living cultures. NucSpot 470 has an excitation/emission of 460/546 nm and can be imaged using standard settings for FITC. Properties: Ex/Em= 460/546 nm (with DNA); Store at 4°C and protect from light.

Hoechst 33528 is slightly more water soluble than Hoechst 33342. The dyes can be used to stain live or fixed cells in buffer or medium at 1 ug/mL, with no wash step required. Properties: Yellow solid; Store dessicated at -20°C, protect from light; Ex/Em= 352/461 nm (with DNA); C25H27Cl3N6O; MW: 623.96; [23491-45-4].

Learn More
A Nucleolus is one of the structures forming a nucleus and becoming the starting point of the ribosomal biogenesis. There are many ribosomal RNA (rRNA) in the nucleolus and transcription of rRNA and processing are carried out. Whereas the morphological change of nucleolus is known as one of the indicators of cancer diagnosis, there have been some scientific reports describing the relation between nucleolus and DNA damage, autophagy, virus infection, and cellular senescence. Nucleolus Bright dyes are small molecules and they bind to RNA in the nucleolus to emit fluorescence. The nucleolus can be observed without any washing steps after staining with Nucleolus Bright dyes. We recommend to co-stain with DAPI in order to image the nucleolus clearly. Modern publications with a high number of citations summarize the various features of cellular senescence on the following page (https://www.dojindo.com/senescent-cell-detection-selection-guide/).
