Compared to the earlier Basal Medium Eagle (BME), MEM is formulated with increased concentrations of amino acids. Because MEM media lacks proteins, lipids, or growth factors, it commonly requires supplementation with fetal bovine serum (FBS).

The medium contains tryptone as a source of nitrogen and amino acids, bile salts act as a selective against Gram-positive organisms and agar is the gelling agent. 5-bromo-4-chloro-3-indolyl-β-D-glucuronide (X-glucuronide) is a chromogen for the detection of the β-D-glucuronidase enzyme which is present in most E. coli* but not other coliforms.*96-97% of E.coli are β-D-glucuronidase positive. The production of hydrogen sulfide, indole formation, and motility are useful diagnostic tests in the identification of Enterobacteriaceae, especially Salmonella spp. and Shigella spp. In 1940, Sulkin and Willett demonstrated motility, hydrogen sulfide production, and carbohydrate fermentation by members of Salmonella and Shigella groups. They called attention to the “brush-like growth” or motility of the typhoid organisms. Brilliant Green Agar is not intended for use in the diagnosis of disease or other conditions in humans.Kristensen, Lester, and Jurgens first described the use of Brilliant Green Agar as a primary plating medium for the isolation of Salmonella. The report described the medium as useful for the differentiation of “paratyphoid B” from other intestinal gram-negative bacilli. Kaufmann modified their formula and used Brilliant Green Agar in addition to Tetrathionate Broth for the isolation of Salmonella from stool specimens. The outstanding selectivity of this medium permits use of moderately heavy inocula, which should be evenly distributed over the surface. Brilliant Green Agar is used in the microbial limits test.
