McCoy's 5A Media (modified) was originally developed by McCoy and his co-workers in 1959 and was known then as Basal Medium 5A. McCoy published the research he had conducted regarding the Basal Medium 5A, which detailed the amino acid requirements for the culture of Novikoff Hepatoma Cells. Basal Medium 5A was subsequently modified resulting in a similar media known as McCoy's 5A Media. Iwakata and Grace further modified the medium for use in work with human myeloblasts. Bioworld Iscove's DMEM w/o Phenol Red, 500 mL
Guilbert and Iscove published their modifications to Dulbecco's Modified Eagle's Medium (DMEM) in 1976. Their modification included the supplements of selenium, additional vitamins and amino acids, sodium pyruvate and HEPES buffer. In addition, ferric nitrate was replaced with potassium nitrate. When further supplemented with albumin, lecithin and transferrin, this medium supported the growth of precursor cell of macrophages and erythocytes without serum supplementation. Iscove's Medium has since been shown to support murine B lymphocytes, hemopoietic tissue from bone marrow, B cells stimulated with lipopolysaccharide, T lymphocytes and a variety of hybrid cells.
