NEOGEN Brilliant Green Agar (Modified), 5 kg
Brilliant Green Agar is used for the selection and differentiation of Salmonella (other than S. typhi) from foods or feedstuffs. Brilliant Green Agar is not intended for use in the diagnosis of disease or other conditions in humans. First introduced by Kristensen et al in 1925 as a selective medium for the isolation of Salmonella (except S. typhi). The medium was modified by the Netherlands Institute for Public Health, Utrecht. The modification was to increase the selectivity of the medium by increasing the dye concentration. This formulation is quoted by the International Standards Organization, standard European Community Methods, the American Public Health Association and the Association of Official Analytical Chemists. The medium is suitable for subcultures from selective enrichment media. MYP Agar Base is not intended for use in the diagnosis of disease or other conditions in humans.Formulated to ISO 7932:2004 MYP was introduced by Mossel and his co-workers in 1967 for the enumeration of Bacillus cereus in foods and was shown to be the most effective for this purpose by Inal in 1972. B. cereus is differentiated from other members of the Bacillus group by two reactions: mannitol fermentation and lecithinase production. Mannitol fermentation on this medium produces a yellow color. B. cereus is mannitol negative and produces red colonies. Orange Serum Agar is used for the cultivation of aciduric microorganisms associated with spoilage of products and is not intended for use in the diagnosis of disease or other conditions in humans.The low pH of fruit juices makes citrus fruit products susceptible to spoilage by yeasts, molds, and the bacteria Lactobacillus and Leuconostoc. In the 1950’s, Hays investigated spoilage in frozen concentrated orange juice. He found that an agar medium containing orange serum (juice) was superior to Lindegren Agar in isolating the microorganisms responsible for spoilage causing a buttermilk off-odor. Murdock, Folinazzo, and Troy found Orange Serum Agar, pH 5.4 to be a suitable medium for growing Leuconostoc, Lactobacillus, and yeasts. NEOGEN YM Broth, 5 kg
YM Broth is used for the cultivation of fungi in a laboratory setting. YM Broth is not intended for use in the diagnosis of disease or other conditions in humans. YM Broth is prepared according to the formulation published by Wickerham. Wickerham suggested that YM Broth, acidified to pH 3.0 – 4.0, be used as an enrichment medium for yeasts from populations also containing bacteria and molds. To favor isolation of fermentative species, add a layer of sterile paraffin oil 1 cm deep on the surface of the inoculated broth. Cultures should be incubated until growth appears, and then streaked onto YM Agar to obtain isolated yeast colonies. To isolate fermentative and oxidative strains, place acidified inoculated YM Broth on a rotary shaker for 1 to 2 days. This favors yeast recovery while preventing sporulation of molds. LB Broth, Miller is a nutritionally rich medium designed for growth of pure cultures of recombinant strains. E. coli is grown to late log phase in LB Medium. Some plasmid vectors replicate to high copy numbers without selective amplification. Some vectors do not replicate so freely and need to be selectively amplified. Chloramphenicol can be added to inhibit host synthesis and prevent replication of the bacterial chromosome. Bile salts and crystal violet are used to inhibit Gram positive and non-enteric organisms. The rich peptone mixture is enhanced by the use of corn starch to absorb toxic metabolites and a buffering system is used to maintain neutral pH. The medium is made selective by the use of various antibiotic cocktails. Thayer and Martin originally recommended the use of vancomycin, colistin and nystatin and the addition of trimethoprim which is useful in preventing the swarming of proteus. Shahidi-Ferguson Perfringens (SFP) Agar is based on the formula by Shahidi and Ferguson and incorporates the supplements, Polymyxin B Sulfate and Kanamycin. An osmotically controlled solution which is an alternative to, and a replacement for, 1/4 strength Ringer’s Tablets (NCM0191K). The presence of a low level of peptone lessens the physiological shock normally experienced by bacterial cells when they are introduced to a diluent such as Ringer’s Solution. The level of peptone is such that multiplication of the organisms is not possible in the time in which the sample will be present in the diluent (1-2 hours).
