The PathHunter U2OS SMO(W535L) MEM-EA Pharmacotrafficking Assay is a stable, engineered cell line for use in studying pharmacochaperones targeting SMO. This cell line enables assessment of chaperone-based stabilization of SMO(W535L) via detection of rescued membrane trafficking. The included cell line overexpresses ProLink-tagged mutant SMO(W535L), which is misfolded and retained in the ER. The cell line also expresses membrane-localized EA. When a pharmacochaperone stabilizes the mutant SMO, it is trafficked to the membrane, producing EFC signal. This stable modified cell line is provided as two vials of cryopreserved cells. The cell line included overexpresses ACVR1B target protein fused to PK alongside BMPR2 target protein fused to EA.
