Deoxyribonuclease I (DNase I) is an endonuclease consisting of a single glycosylated polypeptide chain with two disulfide bonds. DNase is often included in tissue dissociation protocols to digest DNA that has leaked into the dissociation medium as a result of cell damage. DNase I preferentially cleaves phosphodiester linkages adjacent to pyrimidine nucleotides in both single- and double-stranded DNA, yielding polynucleotides with 5’-phosphate and 3’-hydroxyl groups (Bernardi et al.). DNase I has been used for the digestion of human tissues such as microglia (Klegeris & McGeer), cartilage (Dunham & Koch), colon (Fukushima & Fiocchi), epithelium (Fukushima & Fiocchi), liver (Vatakis et al.), lung (Fujino et al.), neural (Fuja et al.), and stem cells (Kusuma et al.).

Protease S. aureus V8 (Endoproteinase-Glu-C) specifically cleaves peptide bonds on the COOH-terminal side of either aspartic or glutamic acids. In the presence of ammonium the enzyme specificity is limited to glutamic sites. It has a molecular weight of 27 000 daltons and optimum pH's of 4.0 and 7.8 with hemoglobin as the substrate. Protease S. aureus V8 is inhibited by diisopropylfluorophosphate and monovalent anions such as F- Cl- CH3COO- and NO3. Enzyme activity is determined by the casein digestion assay. Protease S. aureus is ideal for investigators involved in enzyme and infectious diease research.

Summary: L-Asparaginase is an enzyme that depletes L-Asparagine, an important nutrient for cancer cells, resulting in cancer/tumor cell starvation. The rationale behind asparaginase is that it takes advantage of the fact that acute lymphoblastic leukemia cells and some other suspected tumor cells are unable to synthesize the non-essential amino acid asparagine, whereas normal cells are able to make their own asparagine; thus leukemic cells depend on circulating asparagine. Asparaginase, however, catalyzes the conversion of L-asparagine to aspartic acid and ammonia. This deprives the leukemic cell of circulating asparagine, which leads to cell death.
