Recognizes a 67 kDa glycoprotein, which is identified as CD33. It is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It belongs to the immunoreceptor tyrosine-based inhibitory motif (ITIM)-containing molecules able of recruiting protein tyrosine phosphatases SHP-1 and SHP-2 to signal assemblies; these ITIMs are also used for ubiquitin-mediated removal of the receptor from the cell surface. CD33 is expressed on cells of myelomonocytic lineage, binds sialic acid residues in N- and O-glycans on cell surfaces, and is a therapeutic target for acute myeloid leukemia. Recognizes a protein of 40 kDa, identified as CD7 (also known as gp40, Leu9). CD7 is a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. This MAb recognizes extracellular epitope 2 within the N-terminal Ig domain of human CD147. It is expressed more intensely on thymocytes than on mature peripheral blood T cells. It stimulates the production of interstitial collagenase, gelatinase A, stromelysin-1 and various metalloproteinases (MMPs) by fibroblasts. Recognizes a protein of 33-55 kDa, identified as CD37 (Workshop V; Code CD37.7). CD37 is strongly expressed on normal and neoplastic mature (sIg ) B-lymphocytes. In B-cell ontogeny, CD37 appears after the pre-B-cell stage, is maintained during peripheral B-cell development and is lost upon terminal differentiation into plasma cells.1 CD37 is also present, at low densities, on resting and activated T cells, neutrophils, monocytes, and some myelomonocytic leukemia cells. Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. This antibody recognizes a protein of 21 kDa, identified as the Bax protein. This MAb is highly specific to Bax and shows no cross-reaction with Bcl-2 or Bcl-X protein. Recognizes a protein of 210-220 kDa, which is identified as the complement receptor 1 (CR1)/CD35. This MAb is specific for a site in CR1 that is distal from the C3b-binding site, so that it is unable to block CR1 activity. This MAb is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death.
