This detection kit is a robust, sensitive and easy-to-use assay to determine GPCR activity through detection of intracellular cAMP levels. The HitHunter cAMP assay is intended for use with cells expressing Gαs- or Gαi-coupled GPCRs. This product uses EFC technology, and contains ED-tagged cAMP, anti-cAMP antibody, and pure EA. Upon application to a cell which contains endogenous, untagged cAMP, the anti-cAMP antibody will be sequestered by binding to this untagged cAMP, allowing for EFC between the tagged cAMP and soluble EA. This will produce a proportional level of chemiluminescent signal. This kit format is intended for Biologic drugs with a protocol optimized for use in a 96-well plate. Chemotaxis, a type of cell migration, is a highly integrated, multistep process that orchestrates embryonic morphogenesis, tissue repair and regeneration. The initial response of a cell to a migration-promoting agent is to polarize and extend protrusions in the direction of the attractant; these protrusions can consist of large, broad lamellipodia or spike-like filopodia that are driven by actin polymerization and can be stabilized by extracellular matrix (ECM) adhesion or cell-cell interactions. The CytoSelect™ Cell Migration Assay Kit utilizes polycarbonate membrane inserts (8 µm pore size) to assay the migratory properties of cells. The kit contains 96 polycarbonate membrane inserts with 3 um pores in a 96-well plate, through which migratory cells are able to pass. Cells are quantified using the provided fluorometric dye. The 3 µm pore size is optimal for measuring the migration of leukocytes.
