The Cell Proliferation Tracer Kit (Fluorometric, Violet 450) provides a robust method for long-term cell labeling and monitoring cell division both in vivo and in vitro. The dye passively diffuses into live cells, where it is converted into a fluorescent form by intracellular esterases and covalently linked to amine groups on proteins for intracellular retention. This enables the tracking of multiple cell generations by flow cytometry through the principle of dye dilution, where fluorescent signal partitions equally between daughter cells during M phase, resulting in progressively dimmer peaks. The kit is compatible with fixation and permeabilization.

An antibody mediated hemagglutination control is useful as a qualitative index for the integrity of the RBCs in the assay. Further serial dilutions up to 1:400 can be included in the assay if an independent or antibody-based control is required in the assay.

Oxidative stress is defined as an increase in the production of reactive oxygen species (ROS) due to an imbalance between antioxidant and oxidants. Advanced Oxidation Protein Products (AOPP) are uremic toxins created during oxidative stress through the reaction of chlorinated oxidants, such as chloramines and hypochlorous acid, with plasma proteins. AOPPs are structurally similar to Advanced Glycation End-Product (AGE) proteins and exert similar biological activities. The OxiSelect™ AOPP Assay Kit offers a simple, reproducible, and consistent system for the detection of advanced oxidation protein products in plasma, lysates, and tissue homogenates. This kit includes a Chloramine standard and an AOPP Human Serum Albumin conjugate for use as a positive control. The OxiSelect™ In Vitro ROS/RNS Assay Kit is an assay for measuring the total free radical presence of a sample. The assay employs a proprietary quenched fluorogenic probe, DCFH-DiOxyQ), which is based on similar chemistry to the popular DCFH-DA. The DCFH-DiOxyQ probe is first primed with a quench removal reagent, and subsequently stabilized in the highly reactive DCFH form. In this reactive state, ROS and RNS species can react with DCFH, which is rapidly oxidized to the highly fluorescent 2’, 7’-dichlorodihydrofluorescein (DCF) . The DCFH-DiOxyQ probe can react with hydrogen peroxide, peroxyl radical, nitric oxide, and peroxynitrite anion. These free radical molecules are representative of both ROS and RNS, thus allowing for measurement of the total free radical population within a sample. The kit has a detection sensitivity limit of 40 nM for H2O2. Each kit provides sufficient reagents to perform up to 5x96 assays, including standard curve and unknown samples.
