Recombinant TEV protease with a His-tag is supplied in a 5000 Units format for site-specific cleavage of fusion proteins containing a TEV recognition sequence. The enzyme is widely used to remove affinity tags or solubility tags after recombinant protein expression and purification. The His-tag enables removal of TEV protease after digestion by immobilized metal affinity chromatography. This detection kit is a robust, sensitive and easy-to-use assay to determine GPCR activity through detection of intracellular cAMP levels. The HitHunter cAMP assay is intended for use with cells expressing Gαs- or GαI-coupled GPCRs. This product uses EFC technology, and contains ED-tagged cAMP, anti-cAMP antibody, and pure EA. Upon application to a cell which contains endogenous, untagged cAMP, the anti-cAMP antibody will be sequestered by binding to this untagged cAMP, allowing for EFC between the tagged cAMP and soluble EA. This will produce a proportional level of chemiluminescent signal. This kit format is intended for Biologic drugs with a protocol optimized for use in a 96-well plate.
