Coelenterazines are luminescent enzyme substrates for apoaequorin and Renilla luciferase. Apoaequorin is used as a calcium indicator whereas Renilla luciferase is commonly used as a reporter of transcription regulation. Coelenterazine binds apoaequorin to form an aequorin complex which emits light upon calcium binding, and can measure a broad range of calcium concentrations from ~0.1 uM to >100 uM. Renilla luciferase is widely used as a reporter protein and as a donor in bioluminescence resonance energy transfer (BRET) to study protein-protein interactions. Biotium Coelenterazine E
Coelenterazine e is a synthetic derivative of coelenterazine. It is a superior substrate for Renilla luciferase generating nearly 40% more light, as well as has an initial light intensity more than 7 times higher than native coelenterazine. Coelenterazine e/Renilla luciferase has two emission peaks centered at 418 and 475 nm, respectively. Coelenterazine e is also a good substrate for apoaequorin compared with native coelenterazine. The e-aequorin compelx has a faster luminescence rise time and shows two emission peaks at 405 and 465 nm respectively, with the ratio of the peak heights dependent on calcium concentration in the range pCa 5-7. Peroxidase reacts with the TMB substrate solution to produce a soluble blue reaction product that can be read at 370 nm or 620-650 nm for ELISA applications. Stop solutions can be added to prevent further color development and reading the absorbance values at the desired wavelengths.
