Concanavalin A (Con A) is one of the most widely used and well characterized lectins. Con A has broad applicability primarily because it recognizes a commonly occurring sugar structure, a-linked mannose. Agarose Bound Con A is prepared using affinity-purified lectins and heat stable, cross-linked 4% agarose beads. The attachment of the lectins to the beads through a hydrophilic spacer arm is carefully controlled to preserve lectin activity and minimize conformational changes of the bound lectins. The recommended inhibiting/eluting sugar is a mixture of 200 mM a-methylmannoside/200 mM a-methylglucoside.

The disaccharide galactosyl (α-1,3) N-acetylgalactosamine is also a potent inhibitor of this lectin.Fluorescein labeled Vicia villosa lectin has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated fluorochromes. The excitation maximum is at 495 nm and the emission maximum is at 515 nm.
