3-D Life is the most flexible system for biomimetic hydrogel design. It uses two polymers that form a hydrogel by covalent crosslinking at ambient temperature. Chemical reaction is non-toxic to cells and occurs rapidly, preventing cell settlement before gel is formed. 3-D Life Dextran-CD Hydrogel SG Kit provides reagents to set up slow gelling, cell-compatible hydrogels for 3D cell cultures. Thiol groups on crosslinker CD-Link form stable thioether bonds with thiol-reactive groups on SG-Dextran, resulting in gel formation. Mixed at physiological pH 7.2 for optimal cell compatibility. CD-Link is designed for MMP cleavage, allowing cell spreading/migration if they express the indicated MMPs. Cell adhesion peptides can be attached to SG-Dextran to provide a cell-adhesive matrix. Dextran hydrogels can be dissolved by adding dextranase, allowing easy cell recovery. One kit allows formation of up to 2 ml hydrogel depending on gel stiffness. Advanced Biomatrix HyStem Version 2.0 Thiol-Modified Hyaluronan Hydrogel Kit, 7.5 mL
HyStem contains thiolated hyaluronic acid for easy 3D cell culture. HyStem version 2.0 is reconstituted with a buffered 1X PBS. The resulting components will also result in a pH neutral isotonic salt hydrogel. The ending pH, osmo, and rheological properties are the same from Version 1.0 to Version 2.0. HyStem hydrogels are xeno-free and completely customizable. The hydrogel is formed when the crosslinking agent, Extralink-Lite (PEGDA) is added to a mixture of Glycosil (thiol-modified hyaluronan). Recommended for applications requiring specific attachment factor optimization as well as non-adherent cell culture, cellular applications. Extracellular matrix proteins can be mixed into the hydrogel and incorporated non-covalently before gelation. The HyStem Hydrogel Kits are optimal for culturing stem cells whose natural environment is rich in hyaluronic acid (HA).
