ASP-8731 is an orally active BACH1 inhibitor that activates antioxidant, anti-inflammatory, and globin gene pathways by relieving BACH1's inhibitory effect on NRF2-mediated gene transcription. It upregulates HMOX1, FTH1, and various globins (HGB, HBG, HBA), increasing fetal hemoglobin (HbF) and inducing F-cell production. It also reduces inflammatory responses, white blood cell counts, heme-induced glutathione depletion, and microcirculatory stasis, showing potential for inhibiting sickle cell disease and related hematological disorders.
