The 10X Transfer Buffer-Electro Blotting is the most commonly used buffer in the wet transfer system for the electrotransfer of proteins from SDS-PAGE gels to nitrocellulose or PVDF membranes. The addition of 10% methanol in thhis buffer prevents the gel from swelling during transfer and enhances efficiency of low abundancy proteins and protein binding to nitrocellulose membrane. This buffer is also used as a running buffer for native protein gel electrophoresis.

The contained desthiobiotin is a modified form of biotin that displaces Strep-tagII and Twin-Strep-tag fusion proteins from the modified biotin-binding pocket of Strep-Tactin in a competitive manner, resulting in a mild elution.
