New England Biolabs, Inc. T4 PDG (T4 Endonuclease V) – 2000 units
T4 PDG (pyrimidine dimer glycosylase) has both DNA glycosylase and AP lyase activity. The 16 kd protein recognizes cis-syn-cyclobutane pyrimidine dimers caused by UV irradiation. The enzyme cleaves the glycosyl bond of the 5' end of the pyrimidine dimer and the endonucleolytic activity cleaves the phosphodiester bond at the AP site.

A high fidelity restriction endonuclease that recognizes the sequence G^TCGAC. High Fidelity (HF) Restriction Enzymes have 100% activity in rCutSmart Buffer; single-buffer simplicity means more straightforward and streamlined sample processing. HF enzymes also exhibit dramatically reduced star activity. HF enzymes are all Time-Saver qualified and can therefore cut substrate DNA in 5-15 with the flexibility to digest overnight without degradation to DNA. Engineered with performance in mind, HF restriction enzymes are fully active under a broader range of conditions, minimizing off-target products, while offering flexibility in experimental design. New England Biolabs, Inc. Micrococcal Nuclease – 320000 gel units
Micrococcal nuclease is derived from Staphylococcus aureus and is a relatively non-specific endo-exonuclease. It is purified from a recombinant E. coli strain that digests double-stranded, single-stranded, circular and linear nucleic acids. The enzyme is active in the pH range of 7.0 - 10.0, with optimal activity at pH 9.2 for both RNA and DNA substrates. Cleavage preferences have been observed at sites rich in adenylate, deoxyadenylate or thymidylate. Both DNA and RNA are degraded to 3 phosphomononucleotides and dinucleotides. Micrococcal Nuclease works well with the SimpleChIP Enzymatic Chromatin IP Kit (Agarose Beads) from Cell Signaling Technology.

New England Biolabs, Inc. PreCR Repair Mix - 30 reactions
The PreCR Repair Mix is an enzyme cocktail formulated to repair damaged template DNA prior to its use in the polymerase chain reaction (PCR), microarrays or other DNA technologies. PreCR is active on a broad range of DNA damages, including those that block PCR (e.g. apurinic/apyrimidinic sites, thymine dimers, nicks and gaps) and those that are mutagenic (e.g. deaminated cytosine and 8-oxo-guanine). In addition, it will remove a variety of moieties from the 3end of DNA leaving a hydroxyl group. The PreCR Repair Mix will not repair all damages that inhibit/interfere with PCR. For example, it will not repair 8-oxo-7,8-dihydro-2'deoxyadenosines or fragmented DNA. In fact, the ligase present in the mix is very active at sealing nicks in DNA but does not ligate blunt ends or nicks near a mismatch effectively. This limits the possibility of chimeric gene formation as a product of ligation. The PreCR Repair Mix can be used in conjunction with any thermophilic polymerase.
New England Biolabs, Inc. RNase Inhibitor, Human Placenta – 10000 units
RNase Inhibitor, Human Placenta is a recombinant human placental protein which specifically inhibits ribonucleases (RNases) A, B and C. It is not effective against RNase 1, RNase T1, S1 Nuclease, RNase H or RNase from Aspergillus. In addition, no inhibition of polymerase activity is observed when RNase Inhibitor, Human Placenta is used with Taq DNA Polymerase, AMV or M-MuLV Reverse Transcriptases, or Phage RNA Polymerases (SP6, T7, or T3). The 50 kDa protein inhibits RNases by binding noncovalently in a 1:1 ratio with an association constant greater than 1014.

T4 RNA Ligase 2' truncated (T4 Rnl2'truncated) specifically ligates the pre-adenylated 5' end of DNA or RNA to the 3 end of RNA. The enzyme does not require ATP for ligation but does need the pre-adenylated substrate. T4 Rnl2'truncated is expressed from a plasmid in E. coli which encodes the first 2'9 amino acids of the full length T4 RNA Ligase 2' Unlike the full length ligase, T4 Rnl2'truncated is unable to adenylate the 5' end of the substrate, and as a result it cannot ligate the phosphorylated 5' end of RNA or DNA to the 3' end of RNA . This enzyme, also known as Rnl2'(1-2'9) has been used for optimized linker ligation for the cloning of microRNAs. This enzyme reduces background ligation because it can only use adenylated primers).
