DNase I (RNase-free) is an endonuclease that digests single- or double- stranded DNA, generating dinucleotide, trinucleotide, and oligonucleotide products with 5´-phosphorylated and 3´-hydroxylated ends. The activity of DNase I (RNase-free) dependent on Ca2+ and can be activated by Mg2+ or Mn2+. In the presence of Mg2+, DNase I (RNase-free) can randomly cleaves arbitrary sites of double-stranded DNA; in the presence of Mn2+, DNase I (RNase-free) can simultaneously recognize both strands of DNA and cleaves at nearly same sites. DNase I (RNase-free) can act on single stranded DNA, double stranded DNA, chromatin, and RNA: DNA hybrid strands. RNase Inhibitor, Murine (Glycerol-free) is a 50 kDa recombinant protein of murine origin. The inhibitor specifically inhibits RNases A, B and C by binding noncovalently in a 1:1 ratio with high affinity. It is not effective against RNase 1, RNase T1, S1 Nuclease, RNase H or RNase from Aspergillus. In addition, no inhibition of polymerase activity is observed when RNase Inhibitor is used with Taq DNA Polymerase, AMV or M-MuLV Reverse Transcriptases, or Phage RNA Polymerases (SP6, T7, or T3). Recombinant murine RNase inhibitor lacks the cysteines identified in the human version to be sensitive to oxidation, which causes inactivation of the inhibitor. As a result, RNase Inhibitor, Murine has significantly improved resistance to oxidation compared to the human/porcine RNase inhibitors, and is stable at low reducing agent concentrations (≤ 1 mM). This makes it ideal for reactions where low concentration reducing agent is required (e.g. Real-time RT-PCR).
