Cross reacting material (CRM197) is a nontoxic variant of diphtheria toxin with a single mutation (resulting in a Glycine to Glutamate substitution at postion 52). CRM functions as a carrier protein for polysaccharides and haptens making them immunogenic. CRM is widely used in polysaccharde conjugate vaccines. CRM197 has the advantage of being a well defined protein in contrast to formaldehyde treated toxin (toxoid) which is non-specifically cross linked and subject to rearrangement and loss of epitopes. Tetanus toxin is a potent neurotoxin (150,000 Da) produced by the anaerobic bacterium Clostridium tetani.
