Recognizes a protein of 205 kDa-220 kDa, identified as CD45RA (Workshop III). CD45RA is isoforms of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4 T-cells, 50% of peripheral CD8 T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. Recombinant protein. Human asprosin aa 2732-2871 is fused at the C-terminus to a His-tag. Source E. coli. Lyophilized from 0.2µm-filtered solution in 50mM Tris-HClpH 8.2, 200mM NaCl. Asprosin is a new fasting-induced protein hormone that targets the liver to increase plasma glucose levels. Asprosin is the C-terminal cleavage product of the protein pro-Fibrillin-1. Asprosin is secreted from white adipose tissue and increases hepatic glucose production by using cAMP as a second messenger, leading to activation of protein Kinase A. Reduction of Asprosin levels protects against metabolic syndrome-associated hyperinsulinism. Asprosin may act as a circulating hunger signal. Indeed, peripherally injected recombinant asprosin can cross the blood-brain barrier and intracerebroventricular i.c.v. injection of recombinant asprosin stimulated appetite in wild-type mice, indicating a central mechanism of action.

The first step in successful chromatin immunoprecipitation is shearing the chromatin into 200-1000 bp fragments. This has traditionally been performed by subjecting the isolated chromatin to different pulses of sonication. Sonication is an effective method for shearing DNA, and is the most widely-used method. In some circumstances, however, it can be difficult to optimize due to complications arising from emulsification and overheating. Because of this, Active Motif has developed a robust and user-friendly method to shear chromatin by enzymatic treatment. The Enzymatic Shearing Kit uses a proprietary Enzymatic Shearing Cocktail that quickly shears DNA into 200-1000 bp fragments. Because enzymatic shearing is solely time and temperature dependent, the problems associated with sonication are eliminated and ChIP results are improved.
