Biotium 10-Acetyl-3, 7-dihydroxyphenoxazine
10-Acetyl-3,7-dihydroxyphenoxazine (N-Acetyl-3,7-dihydroxyphenoxazine or ADHP), marketed under the name Amplex Red, is regarded as the best fluorogenic substrate for peroxidase because it is highly specific and stable. The substrate itself is nearly colorless and nonfluorescent until it is oxidized by H2O2 (reacting in a 1:1 stoichiometry) in the presence of horseradish peroxidase (HRP) to become the highly red fluorescent resorufin. H2O2 is produced by many enzymatic reactions, and therefore ADHP can be used to detect the activity of many different enzymes. Digoxigenin-UTP can replace UTP in reactions catalyzed by T3, T7 or SP6 RNA polymerases. The digoxigenin-labeled RNA transcripts produced by these reactions are suitable for a wide range of applications such as nucleic acid hybridization, sequencing, and genome analysis. The transcription reaction produces multiple RNA copies of the DNA template(s) during a short incubation period. RNA probes offer higher target specificity and greater sensitivity than the corresponding DNA-DNA hybrids. The single-stranded RNA probes offer selectivity unavailable with double-stranded DNA probes, because they are strand-specific. Furthermore, RNA probes hybridize much more efficiently to target molecules than DNA probes because there is no self-hybridization. Alternative name: Digoxigenin-11-UTP. Concentration: 10mM. Diaminobenzidine (DAB)is the most commonly used substrate for the histochemical detection of peroxidase. It reacts with HRP to produce an insoluble brown precipitate and can be used in staining tissues and on membranes. The main disadvantage of DAB is that it must be used immediatley to avoid reaction with the enzyme.
