The OneTaq One-Step RT-PCR Kit offers sensitive and robust end-point detection of RNA templates. cDNA synthesis and PCR amplification steps are performed in a single reaction using gene-specific primers, resulting in a streamlined RT-PCR protocol. The kit combines three optimized mixes: OneTaq One-Step Enzyme Mix, OneTaq One-Step Reaction Mix and OneTaq One-Step Quick-Load Reaction Mix. The Enzyme Mix combines ProtoScript II Reverse Transcriptase, Murine RNase Inhibitor and OneTaq Hot Start DNA Polymerase. ProtoScript II Reverse Transcriptase is a mutant M-MuLV reverse transcriptase with reduced RNase H activity and increased thermostability. In the Luna One-Step RT-qPCR Kit, Hot Start Taq DNA Polymerase is combined with a novel WarmStart-activated reverse transcriptase, allowing dual control of enzyme activity via reversible, aptamer-based inhibition. This temperature-dependent activation helps to prevent undesirable non-specific priming and extension prior to thermocycling, providing added security for setting up reactions at room temperature. The engineered WarmStart Luna Reverse Transcriptase also possesses higher thermostability than many other RTs, allowing an optimal reaction temperature of 55°C. For difficult targets/templates, higher RT step temperatures of up to 60°C can be used without compromising Luna performance. Note that to ensure full activation of the WarmStart Luna RT, incubation at temperatures lower than 50°C is not recommended.

The unique lysis buffer rapidly lyses cells from trace samples and inactivates cell RNA enzymes. The lysate mixture passes through a gDNA Remove Column, where gDNA is removed and RNA is filtered through. After adjusting the binding conditions by ethanol, RNA is selectively bound to the silicon membrane of the adsorption column in the state of high disordered salt. In a series of rapid washing-centrifugation steps, the purified RNA is eluted from the silicon membrane with low salt RNase-free H2O.
