DMEM,with High Glucose, L-glutamine, Sodium Pyruvate and, 25mM HEPES Dulbecco's Modified Eagle's Medium (DMEM) is a modification of Basal Medium Eagle (BME), containing four-fold concentrations of the amino acids and vitamins found in BME, as well as additional supplementary components. The original formulation of DMEM contained 1000 mg/L of glucose, but has since been raised in some modifications to 4500 mg/L, giving the terms "Low Glucose" and "High Glucose". Several other modifications over the years, in addition to the alternative glucose levels, have broadened the spectrum of cell types supported by DMEM

In the 1950s it was found that mammalian cells do not only need the 10 essential amino acids, but also cystine, tyrosine and glutamine. In addition to these three amino acids, BME includes also eight B-vitamins. BME was originally used for the cultivation of murine L-cells and HeLa cells. Along with the cultivation of normal mammalian cells BME is very suitable for transformed cells. BME is the precursor to other common media such as MEM, DMEM, and Glasgow's Medium.

Early attempts to cultivate normal mammalian fibroblasts and certain subtypes of HeLa cells revealed they had specific nutritional requirements that could not be met by Eagles Basal Medium (BME). Subsequent studies using these and other cells in culture indicated additions to BME could be made to aid growth of a wider variety of fastidious cells.MEM which incorporates these modifications includes higher concentrations of amino acids so the medium more closely approximates the protein composition of cultured mammalian cells. MEM has been used for cultivation of a wide variety of cells grown in monolayers. Optional supplementation of non-essential amino acids to the formulations that incorporate either Hanks or Earles salts has broadened the usefulness of this medium.
