With excitation/emission maxima of 495/515 nm, DAF-FM can be detected using any instrument suitable for detecting fluorescein.
AdipoGen 7-Azido-4-methylcoumarin
Chemical. CAS 95633-27-5. Formula C10H7N3O2. MW 201.18. Synthetic 7-Azido-4-methylcoumarin is a highly sensitive and selective fluorogenic H2S probe. The aromatic azide moiety of AzMC is selectively reduced in the presence of H2S, producing the fluorescent 7-amino-4-methylcoumarin AMC with a concomitant increase in fluorescence with lambdaex = 365 nm and lambdaem = 450 nm. It is a photoaffinity labeling probe for the substrate binding site of human sulfotransferase 1A1 SULT1A1. 7-Azido-4-methylcoumarin is a tool for monitoring the activity of pyridoxal-5-phosphate PLP-dependent enzymes e.g. cystathionine beta-synthase CBS, cystathionine gamma-lyase CGL and tryptophan synthase TS and to identify novel cystathionine beta-synthase CBS inhibitors and activators.

AdipoGen BCECF-AM
Chemical. CAS 117464-70-7. Formula C42H40O21. MW 808.69. Synthetic. Widely-used fluorescent indicator for estimating intracellular pH. This membrane-permeant compound is modified by intracellular esterases to produce BCECF, a polar fluorescein derivative that is retained by cells. Intracellular BCECF is stable and has an efflux half-life of greater than two hours. Intracellular pH measurements are made by ratio imaging, which involves determining the pH-dependent ratio of emission intensity detected at 535nm when the dye is excited at 490nm vs. the emission intensity when excited at 440nm. This approach is amenable to either spectrofluorometry or flow cytometry. BCECF-AM ester can also be used to investigate intracellular changes in other ions, including potassium. Spectral properties lambdaex 490nm lambdaem 535nm lambdaex=505nm lambdaem=520nm in DMSO.
