The NUCLEAR-ID Green Chromatin Condensation Detection Kit detects nuclear condensation using a DNA intercalating dye that brightly stains the condensed chromatin of apoptotic cells, but only dimly stains the uncondensed chromatin of healthy living cells. Mechanism of Action The cell-permeable dye used in this application is an aromatic, planar cationic structure that inserts between stacked base pairs on the DNA duplex, providing an environmentally-dependent fluorescence enhancement of the dye molecules and large increases in fluorescence signal relative to the free dye in solution. Since the signal enhancement provides a proportional response, direct quantitation of DNA is possible. Further signal increase is observed upon DNA condensation during apoptosis.Applications: Flow Cytometry, Fluorescence microscopy, Fluorescent detection .

Annexin V is a cellular protein, which plays important roles in the inhibition of the activity of phospholipase A1 and blood coagulation by competing for phosphatidylserine (PS) binding sites with prothrombin. Annexin V has a high affinity to PS and is used as a probe to detect cells that have expressed PS on the cell surface. After initiating apoptosis, cells translocate phosphatidylserine (PS) from the inner face of the plasma membrane to the cell surface soon, which can be easily detected by Annexin V.Annexin V-Cy5 Apoptosis Detection Kit uses a fluorescent conjugate of Annexin V that can easily detect PS on the cell surface after initiating apoptosis. MW 343.4. m-3M3FBS is a cell permeable and specific activator of phospholipase C PLC that has been shown to stimulate the PLC-mediated intracellular Ca2 release, inositol phosphates production and superoxide generation in various cell types. It activates all PLC isotypes beta2, beta3, gamma1, gamma2 and delta1 in vitro, but not heterotrimeric G proteins, PI 3-kinase or PLD. m-3M3FBS also affects calcium homeostasis independently of PLC activation. m-3M3FBS induces apoptosis in tumor cells through caspase activation, down-regulation of XIAP and intracellular calcium signaling. m-3M3FBS has been shown in vitro to trigger mitochondrial damage and activate NLRP10 a bona fide inflammasome binding to Asc and Caspase-1 in keratinocytes. NLRP10 acts also as inflammasome in intestine epithelial cells and protects against autoinflammation in the gut.

SP600125 is an orally active, reversible and ATP-competitive broad-spectrum JNK inhibitor for JNK1, JNK2 and JNK3 IC50 = 40nM, 40nM and 90nM, respectively. In Jurkat T cells, SP600125 suppressed the phosphorylation of c-Jun with IC50 of 5-10 µM. It has 10-fold greater selectivity against MKK4, 25-fold greater selectivity against MKK3, MKK6, PKB and PKCalpha, and 100-fold selectivity against ERK2, p38, Chk1, EGFR.
