Tryptic Soy Agar with 0.03% Congo Red (TSA w/CR) is a non-selective, differential medium used for the cultivation of a wide variety of non-fastidious microorganisms. Congo red is utilized to stain the cells of fungi and the outer membrane of Gram-negative bacteria. Additionally, studies have indicated virulent Shigella flexneri serotype 2a, grown in the presence of congo red at 37°C, showed increased levels of three proteins with MRS of 43K, 58K, and 63K proteins) in the Sarkosyl-soluble membrane fractions. Baird-Parker (BP) Agar is a selective and differential medium used for the isolation and presumptive identification of coagulase-positive Staphylococcus from clinical specimens, foods, air, water, and other materials. Colonies of S. aureus form a characteristic black pigmentation by reducing the potassium tellurite and form clear zones due to proteolysis of the egg yolk emulsion. Chameleon™ mEI Agar is a selective and differential, chromogenic medium used for the identification of Enterococcus. Levin, et al. developed a two-step membrane filter method to measure enterococci in fresh and marine recreational waters. Using mE Agar, the method required a 48-hour incubation and a transfer of the membrane to another substrate medium, Esculin Iron Agar, to differentiate enterococci. In 1997, the EPA improved the mE Agar formula by reducing the amount of triphenyltetrazolium chloride contained in the medium and also added the chromogen indoxyl-B-D-glucoside. (Wallerstein Laboratory) WL Nutrient (WLN) Agar is a selective and non-differential medium used for the cultivation of yeasts, molds, and bacteria encountered in brewing and industrial fermentation processes. WLN was developed by Green and Gray while studying various fermentation processes. An exhaustive study examining methods of fermentation control procedures in worts, beers, liquid yeasts, and similar fermentation products led to the development of WL Nutrient Medium. At a pH of 5.5, counts of viable baker’s yeast will grow.
