Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. CD28 is also expressed on human fetal NK cells and some NK cell lines, whereas on murine NK cells the CD28 expression is much broader.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). Recognizes a protein of 47-55 kDa, which is identified as FOXP3. Its precise epitope is not known, but it has been mapped to the N-terminal portion of the protein. The FOX family of transcription factors is a large group of proteins that share a common DNA binding domain termed a winged-helix or forkhead domain. During early development, FOXP1 and FOXP2 are expressed abundantly in the lung, with lower levels of expression in neural, intestinal and cardiovascular tissues, where they act as transcription repressors. FOXP1 is widely expressed in adult tissues, while neoplastic cells often exhibit a dramatic change in expression level or localization of FOXP1. Mutations in FOXP3 gene cause IPEX, a fatal, X-linked inherited disorder characterized by immune dysregulation. The FOXP3 protein is essential for normal immune homeostasis. Recognizes a single chain glycoprotein of 70 kDa, identified as CD55 (also known as decay accelerating factor, DAF). CD55/DAF is widely expressed on cells throughout the body including leukocytes, erythrocytes, epithelium, endothelium, and fibroblasts. It is a Glycosyl phosphatidylinositol anchored (GPI-anchored) member of the membrane bound complement regulatory proteins that inhibit autologous complement cascade activation. It prevents the amplification steps of the complement cascade by interfering with the assembly of the C3-convertases, C4b2a and C3bBb, and the C5-convertase, C4b2a3b and C3bBb3b. CD55 also serves as receptor for CD97 and for echovirus and Coxsackie B virus. This antibody is specific to Insulin-like Growth Factor (IGF-1) and shows minimal cross-reaction with IGF-11, Proinsulin, MSF, and Insulin. IGF-1 is a polypeptide growth factor with two isoforms that are produced by alternative splicing. Isoform 1 is also known as IGF-IB while isoform 2 is known as IGF-IA. IGF-1 stimulates the proliferation of a wide range of cell types including muscle, bone and cartilage tissue. It functions as an autocrine regulator of growth. Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. Recognizes a single glycoprotein of 520 kDa, identified as mucin 2 (MUC2). This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC3. Its epitope has been defined as GTQTP (GlyThrGlnThrPro). Mucins are high molecular weight glycoproteins, which constitute the major component of the mucus layer that protects the gastric epithelium. MUC2 is specifically expressed in goblet cells of the small intestine & colon; in about 65% of colonic carcinomas and about 40% of gastric carcinomas. This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). Expression of Bcl-2 inhibits programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of Bcl-2 protein, whereas the normal or hyperplastic germinal centers are negative. CD19 is a transmembrane glycoprotein that contains two extracellular immunoglobulin-like domains. CD19 is present in both benign and malignant B-cells and is considered to be the most reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells, and in scattered cells of the inter-follicular areas. Anti-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However, in contrast to CD20, expression of CD19 is continuous throughout B-cell development and through terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cell neoplasms commonly at a lower intensity than normal B-cell counterparts. Cytokeratin 17 (CK17) is normally expressed in the basal cells of complex epithelia but not in stratified or simple epithelia. Antibody to CK17 is an excellent tool to distinguish myoepithelial cells from luminal epithelium of various glands such as mammary, sweat and salivary. CK17 is expressed in epithelial cells of various origins, such as bronchial epithelial cells and skin appendages. It may be considered asepithelial stem cellmarker because CK17 Ab marks basal cell differentiation. CK17 is expressed in SCLC much higher than in LADC. Eighty-five percent of the triple negative breast carcinomas immunoreact with basal cytokeratins including anti-CK17. Also important is that cases of triple negative breast carcinoma with expression of CK17 show an aggressive clinical course. Recognizes a protein of 33-55 kDa, identified as CD53 (Workshop VI; Code N-L033). It is expressed on monocytes and macrophages, dendritic cells, osteoblasts and osteoclasts, and on T and B cells from every stage of differentiation but is absent from platelets, red blood cells. CD53 appears to be the marker with widest reactivity as well as the marker with the strictest specificity to hematopoietic cells. CD53 is a type III membrane with both termini in the cytoplasm and two loops in the extracellular environment. This molecule, in common with other members of tetraspan family, is involved in cellular activation as part of a signal transduction complex involving other membrane glycoproteins. This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The highly leukemogenic avian retrovirus E26 contains two oncogenes, v-Myb and v-Ets, which are expressed together as a fusion protein. The cellular homolog of v-Myb, designated c-Myb, encodes a transcription factor.
