The EasySep™ Non-Human Primate B Cell Isolation Kit is designed to isolate untouched and highly purified B cells from fresh or previously frozen non-human primate peripheral blood mononuclear cells (PBMCs) in as little as 20 minutes by immunomagnetic negative selection. The EasySep™ procedure involves labeling unwanted cells with antibody complexes and magnetic particles. The magnetically labeled cells are separated from the untouched desired cells by using an EasySep™ magnet and simply pouring or pipetting the desired cells into a new tube. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.This kit has been verified for use with rhesus and cynomolgus macaques.

The EasySep™ Human CD3 Positive Selection Kit II is designed to isolate CD3+ cells from fresh or previously frozen peripheral blood mononuclear cells by positive selection. Desired cells are targeted with Tetrameric Antibody Complexes recognizing CD3 and dextran-coated magnetic particles. The cocktail also contains an antibody to human Fc receptor to minimize nonspecific binding. Labeled cells are separated using an EasySep™ magnet without the use of columns. Cells of interest remain in the tube while unwanted cells are poured off. The CD3 antigen is expressed on all T cells and CD56+ NKT cells.This product replaces the EasySep™ Human CD3 Positive Selection Kit (Catalog #18051) for even faster cell isolations.

Based on protein domains known to possess an affinity for ubiquitin, Tandem Ubiquitin Binding Entities (TUBEs) have been developed for the isolation and identification of ubiquitylated proteins. TUBEs display up to a 1000-fold increase in affinity for polyubiquitin moieties over the single ubiquitin binding associated domain (UBA). In addition, TUBEs display a protective effect on polyubiquitinated proteins, allowing for detection at relatively low abundance. These properties effectively “capture” proteins in their polyubiquitylated state. Biotin-TUBEs allow for the detection of polyubiquitin and polyubiquitylated proteins by ligand blotting (“far Western”) without heating the membrane. This reagent is a superior alternative to traditional polyubiquitin immunodetection techniques, such as anti-ubiquitin IgGs.

Biotin K63-TUBE binds K63-linked polyubiquitin with a 1000 to 10,000-fold preference for K63 over other linkage types and provides a sensitive and cost-effective tool for determining the abundance of K63-linked polyubiquitin in the cell, or identifying the linkage-type associated with a particular protein of interest. Biotin K63-TUBE is useful for detection and characterization of K63-linked polyubiquitinated proteins by Far Western blotting, affinity purification from lysates and biological fluids, and in situ detection. Far Westerns with Biotin K63-TUBE require only minor modifications to immunoblot protocols, the most notable of which is to avoid subjecting the blot to denaturing conditions, e.g. boiling or autoclaving, which are common treatments used in Western blotting protocols for ubiquitin antibodies. Biotin K63-TUBE substitutes for the anti-ubiquitin IgG as the primary detection reagent. Secondary detection is accomplished with an avidin or streptavidin enzyme conjugate.
