3-(3-pyridinyl)-1-(4-pyridinyl)-2-propen-1-one (3PO) is a small-molecule inhibitor of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3) used as a research probe to study glycolysis and cancer cell metabolism. It reduces glycolytic flux and glucose uptake, lowers intracellular fructose-2,6-bisphosphate, lactate, ATP, and NAD+/NADH, and shows cellular activity in the low micromolar range.
