This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein can undergo autoproteolytic processing and activation by the apoptosome, a protein complex of cytochrome c and the apoptotic peptidase activating factor 1, this step is thought to be one of the earliest in the caspase activation cascade. This protein is thought to play a central role in apoptosis and to be a tumor suppressor. Alternative splicing results in multiple transcript variants.

This kit includes two apoptosis markers, NucView 488 Caspase-3 Substrate (green) and CF594 conjugated Annexin (red), for profiling apoptotic cells with respect to two important apoptosis events, caspase-3 activation and phosphatidylserine (PS) translocation, by flow cytometry or fluorescence microscopy.Unlike traditional fluorogenic caspase substrates that inhibit caspase activity or require cell lysis, NucView 488 a novel fluorogenic caspase-3 substrate, is non-inhibitory to caspases, and permits apoptosis detection within individual intact cells, in real-time. The non-fluorescent substrate is cleaved by caspase-3/7 in apoptotic cells releasing a high-affinity DNA dye that migrates to the nucleus and emits bright green fluorescence, allowing caspase-3/7 detection and visualization of apoptotic nuclear morphology.
