Abcam MTT Assay Kit ab211091 is an easy-to-use, non-radioactive, and high-throughput assay for measuring cell proliferation, cell viability and cytotoxicity.
The MTT assay protocol is based on the conversion of water soluble MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) compound to an insoluble formazan product. NB: MTT is also available as free molecule as ab146345 (Thiazolyl blue tetrazolium bromide).Viable cells with active metabolism convert MTT into formazan. Dead cells lose this ability and therefore show no signal. Thus color formation serves as a useful and convenient marker of only the viable cells. The measured absorbance at OD 590 nm is proportional to the number of viable cells.The MTT assay is a measure of the metabolic activity of the cells analyzed; the more metabolic activity in the sample, the higher the signal. This should be considered when interpreting the results of the assay.

One quick assessment of angiogenesis is the measurement of the ability of endothelial cells to form three-dimensional structures (tube formation). Endothelial cells form tubes on collagen or fibrin clot coated dishes after several days. Cell Biolabs’ Endothelial Tube Formation Assay Kit utilizes ECM (extracellular matrix) gel prepared from Engelbreth–Holm–Swarm (EHS) tumor cells. Endothelial tube formation on ECM gel closely mimics the in vivo environment and may be used to test angiogenesis inhibitors before in vivo testing. The Endothelial Tube Formation Assay Kit provides a robust system to assess angiogenesis in vitro. Tube formation can be achieved within 18 hours. Following staining the tube with the provided fluorescence dye, the extent of tube formation, such as average tube length and branch point, can be quantified through imaging software. Each kit provides sufficient quantities to perform up to 50 assays in 96-well plates.

Chemotaxis, a type of cell migration, is a highly integrated, multistep process that orchestrates embryonic morphogenesis, tissue repair and regeneration. The initial response of a cell to a migration-promoting agent is to polarize and extend protrusions in the direction of the attractant; these protrusions can consist of large, broad lamellipodia or spike-like filopodia that are driven by actin polymerization and can be stabilized by extracellular matrix (ECM) adhesion or cell-cell interactions. The CytoSelect™ Cell Migration Assay Kit utilizes polycarbonate membrane inserts (8 µm pore size) to assay the migratory properties of cells. The kit contains twelve 5 um polycarbonate membrane inserts in a 24-well plate, through which migratory cells are able to pass. Cells are quantified using the provided fluorometric dye. The 5 µm pore size is optimal for measuring the migration of monocytes and macrophages.

Fluo-4, pentapotassium is a membrane-impermeant green-fluorescent calcium indicator that can be loaded into cells via microinjection or scrape loading. It is an improved analog of Fluo-3 with the two chlorine substituents replaced by fluorines, resulting in increased fluorescence excitation at 488 nm and higher fluorescence signal. Fluo-4 is essentially nonfluorescent without Ca2+ but upon Ca2+ binding, its fluorescence increases at least 100 times. Fluo-4 has a higher Kd than fura-2 and indo-1, it is more useful for measuring high transient Ca2+ concentration during Ca2+ spikes.
