Useful agent for immunometabolism research. Anticancer reducing tumor sizes in mice and anti-inflammatory agent. Shown to bind the enzyme PKM2 to promote formation of stable, enzymatically-active tetramers, thereby promoting the last rate-limiting step of glycolysis, converting PEP to pyruvate. In addition, it prevents PKM2 translocation to the nucleus resulting in reduced glycolysis and HIF-1alpha activation, causing diminished Warburg effect and IL-1beta production.

Potent mitochondrial ATP synthase ATPases F1F0 inhibitor, consequently leading to inhibition of oxidative phosphorylation OXPHOS. Useful agent for immunometabolism research. Inhibits ATP synthase by blocking its proton channel F0 subunit, which is necessary for oxidative phosphorylation of ADP to ATP energy production, significantly reducing electron flow through the electron transport chain. Anticancer agent. Shown to induce apoptosis/necroptosis in a variety of cell types, as well as autophagy. Shown to activate AMPK. Broad antifungal, antibiotic and nematocidal agent.
AdipoGen Cerulenin
Chemical. CAS 17397-89-6. Formula C12H17NO3. MW 223.3. Isolated from Cephalosporium caerulens. Antibiotic and antifungal compound. Inhibitor of bacterial fatty acid synthesis inhibits FabH, FabB and FabF condensation enzymes. Irreversible fatty acid synthase FAS inhibitor. Inhibits sterol and fatty acid biosynthesis. In fatty acid synthesis, reported to bind in equimolar ratio to beta-keto-acyl-ACP synthase, thus inhibiting protein acylation. In sterol synthesis, inhibits HMG-CoA synthetase activity. Palmitoylation a post-translational lipidation inhibitor. Anticancer agent. Apoptosis inducer in tumor cell lines, with upregulated expression and activity of fatty acid synthase FAS. Shown to induce apoptosis via disruption of the interaction between AIF and Hexokinase II HKII. Necroptosis inhibitor by blocking very long-chain fatty acids VLCFA synthesis and accumulation, which is important in the process of necroptosis RIP1-dependent cell death.
AdipoGen Paeoniflorin
Chemical. CAS 23180-57-6. Formula C23H28O11. MW 480.46. Paeoniflorin has diverse cellular actions, including modulating NMDA and TRPV1 receptors. It is reported to inhibit testosterone synthesis and stimulate aromatase activity. It also reduces inflammatory signaling by inhibiting p38 MAP kinase and blocks pancreatic cancer cell apoptosis by suppressing MMP-2/9 and ERK signaling. It has been described as an adenosine A1 receptor activator with neuroprotective and antidepressant effects, as well as hypoglycemic activity. It can activate adenosine A-1 receptors to increase the translocations of PKC and GLUT4, two major signals for glucose uptake, from cytosol to membrane of the white adipocytes in rats. Recently it also was described as a liver X receptor LXR agonist, that can act as a ligand-activated transcription factor to exhibit antihyperlipidemic and neuroprotective effects.
