Mueller demonstrated the effectiveness of Tetrathionate Broth (supplemented) for enriching typhoid and paratyphoid bacilli while inhibiting coliform organisms. Using modified Mueller’s broth, Kauffmann increased the number of positive isolates. Sabouraud Dextrose Agar (SabDex/SDA) is a selective and non-differential medium used for cultivating pathogenic and commensal fungi and yeasts. It is a modification of Dextrose Agar as described by Sabouraud. The high dextrose concentration and acidic pH of the formula permits selectivity of fungi. Lecithin neutralizes quaternary ammonium compounds and polysorbate 80 neutralizes substituted phenolic disinfectants. D/E (Dey-Engley) Neutralizing Broth without Tween 80 is utilized for the neutralization and testing of antiseptics and disinfectants according to the procedures of Engley and Dey. The broth contains a high concentration of lecithin (7.0g/L) and is opaque in appearance. Therefore, turbidity alone cannot be used as a visual indication of growth and subculture onto a suitable solid media is recommended. Organisms that ferment dextrose will cause a color shift from purple to yellow. Sabouraud Dextrose Agar is an agar medium recommended for the isolation, cultivation and identification of non-pathogenic and pathogenic fungi. This medium does not contain antibiotics or inhibitors and can be used for the cultivation of those fungi which are susceptible to antibiotics.
Dissolve 65.0 grams in 1 liter distilled water. Sterilize by autoclaving for 15 minutes.
