Buffered Charcoal Yeast Extract Agar without L-Cysteine (BCYE w/o Cys) is a non-selective and non-differential medium. It is used in combination with BCYE Agar(s) containing L-cysteine HCl for the presumptive identification of Legionella species. L-cysteine HCl is a required growth factor. Ferric pyrophosphate and alpha-ketoglutarate provide additional nutritional requirements. Activated charcoal decomposes hydrogen peroxide, which is a toxic metabolic biproduct, while also collecting CO2 and modifying surface tension of the media. ACES buffer helps maintain the proper pH for optimal growth. Bile Esculin Azide Agar with Vancomycin and Streptomycin (BEAVS) is a selective and differential medium used for isolation and presumptive identification of group D streptococci, Enterococcus faecalis, and Enterococcus faecium. Group D streptococci and enterococci hydrolyze esculin in the presence of bile, resulting in esculetin and dextrose. Esculetin reacts with ferric ions to form brown-black colonies. Oxgall (bile salts) inhibit most strains of non-group D streptococci and enterococci. The medium is supplemented with 8µg/mL Vancomycin and 100µg/mL Streptomycin. Streptomycin inhibits gram-negative organisms present in background flora.
