The included cell line overexpresses ProLink-tagged C5AR1 and EA-tagged β-Arrestin-2. Activation of C5AR1 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. The PathHunter SIRPa Signaling Bioassay consists of human cells engineered to measure drug potency and detect neutralizing antibodies. Engagement of the receptor by its ligand, or agonist antibodies results in phosphorylation of the receptor’s cytosolic tail. The SH2-domain fused to EA binds the phosphorylated receptor, forcing complementation of ED and EA, resulting in formation of an active β-gal enzyme, which hydrolyzes the substrate to generate a chemiluminescent signal. The bioassay kit contains all reagents needed for the assay including pre-qualified cryopreserved cells, detection reagents, cell plating reagent, positive control agonist, and assay plates. This kit assesses ligand (e.g. GLP-2) based activation of GLP2R GPCR activity via detection of increasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type GLP2R, a naturally Gs coupled GPCR. Cayman Chemical Protein Carbonyl Colorimetric
Cayman’s Protein Carbonyl Colorimetric Assay Kit utilizes the DNPH reaction to measure the protein carbonyl content in plasma, serum, cell lysates, or tissue homogenates in a convenient 96-well format. The amount of protein-hydrazone produced is quantified spectrophotometrically at an absorbance between360-385 nm. The carbonyl content can then be standardized to the protein concentration.

Cayman’s Triglyceride Colorimetric Assay Kit provides a simple, reproducible, and sensitive tool for assaying mono-, di-, and triglycerides in plasma, serum, cell culture extracts, and tissue homogenates. The Triglyceride Colorimetric Assay Kit uses the enzymatic hydrolysis of triglycerides by lipase to produce glycerol and free fatty acids. The glycerol released is subsequently measured by a coupled enzymatic reaction system. The glycerol formed in reaction 1 is phosphorylated to glycerol-3-phosphate in a reaction catalyzed by glycerol kinase. The glycerol-3-phosphate is oxidized by glycerol phosphate oxidase producing dihydroxyacetone phosphate and hydrogen peroxide (H2O2). Peroxidase catalyzes the redox-coupled reaction of H2O2 with 4-aminoantipyrine (4-AAP) and N-ethyl-N-(3-sulfopropyl)-m-anisidine (ESPA), producing a brilliant purple color. The absorbance is measured at 540 nm.

Cayman’s PC Assay provides a specific, sensitive, and convenient method for quantifying PC in plasma or serum. In this assay, PC-Specific PLD hydrolyzes PC to choline and phosphatidic acid. The newly formed choline is then used to generate hydrogen peroxide in a reaction catalyzed by choline oxidase. Finally, with peroxidase as a catalyst, hydrogen peroxide reacts with DAOS and 4-aminoantipyrine to generate a blue dye with an optimal absorption at 595 nm.

The measurement can be finished in 1.5 hours. The detection range is from 78 to 5000 pg/mL. The levels of sample EGFR Kinase activity are parallel to the standard curves obtained using the kit standards linearly. These results indicate that this kit can be used to determine relative mass values for natural EGFR Kinase Activity.
