PARP1 (Poly ADP-ribose Polymerase 1) is a key DNA repair enzyme that detects DNA damage and facilitates repair through PARylation, a process in which PARP1 uses NAD⁺ to add poly (ADP-ribose) chains to itself and other proteins. PARP1 activity recruits DNA repair factors to damaged sites and supports genome stability. PARP1 is frequently overexpressed in several cancers, including breast, ovarian, prostate, and lung cancers, making it an important therapeutic target. Some PARP inhibitors not only block PARP1 catalytic activity but also trap PARP1 on damaged DNA, preventing DNA repair and leading to cancer cell death, particularly in BRCA-mutant tumors.
