NucView 488 Caspase-3 substrate is a novel, fluorogenic substrate for real-time or endpoint detection of apoptosis in live cells. The non-toxic, no-wash substrate is non-inhibitory to caspase-3 unlike other caspase substrates or FLICA assays permitting multiday analysis. Consisting of a high-affinity DNA dye linked to a DEVD caspase-3 substrate moiety, the initially non-fluorescent, cell permeable substrate is cleaved by caspase-3 in apoptotic cells and emits green fluorescence for the detection of both caspase-3 activity and apopototic nuclear morphology, in single-cells. As apoptotic rates and expression of asociated markers varies between cells, even within the same population, single-cell detection is important. Suitable for flow cytometry, fluorescence microscopy, live cell imaging systems, or fluorescence microplate reader, staining is compatible with fixation and permeabilization for immunostaining. NucView 488 is validated in over 100 cell types and 200 publications.

The FLUOR DE LYS-Green substrate offers a highly sensitive and convenient alternative to radiolabeled, acetylated histones or peptide/HPLC methods for the assay of histone deacetylases. The assay procedure has two steps. First, the FLUOR DE LYS-Green Substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity (HeLa nuclear or other extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes the substrate so that, in the second step, treatment with the FLUOR DE LYS Developer (BML-KI105) produces a fluorophore. The FLUOR DE LYS-Green substrate has a higher wavelength excitation and emission than the original FLUOR DE LYS substrate (BML-KI104). This avoids interference by quenching or fluorescence from compounds absorbing and/or emitting in the near UV and blue range. Formulation: 50mM solution in DMSO. Purity: ≥95% (HPLC).
