Biotium 10-Acetyl-3, 7-dihydroxyphenoxazine
10-Acetyl-3,7-dihydroxyphenoxazine (N-Acetyl-3,7-dihydroxyphenoxazine or ADHP), marketed under the name Amplex Red, is regarded as the best fluorogenic substrate for peroxidase because it is highly specific and stable. The substrate itself is nearly colorless and nonfluorescent until it is oxidized by H2O2 (reacting in a 1:1 stoichiometry) in the presence of horseradish peroxidase (HRP) to become the highly red fluorescent resorufin. H2O2 is produced by many enzymatic reactions, and therefore ADHP can be used to detect the activity of many different enzymes. Coelenterazine h (2-(4-Dehydroxy) coelenterazine) is a synthetic derivative of coelenterazine. The luminescence intensity of its aequorin complex is more than 10 times higher than that of native coelenterazine. Coelenterazines are luminescent enzyme substrates for apoaequorin and Renilla luciferase. Apoaequorin is used as a calcium indicator. It binds coelenterazine to form an aequorin complex that emits light upon calcium binding and measures concentrations ~0.1 uM to >100 uM. Renilla luciferase is widely used as a reporter of transcription regulation and as a donor in bioluminescence resonance energy transfer (BRET) to study protein-protein interactions. Coelenterazines are also used for chemiluminescent detection of superoxide anions and peroxynitrite in cells or tissues. NucView Caspase-3 Substrates are novel fluorogenic substrates for real-time or endpoint detection of apoptosis in live cells. The non-toxic, no-wash substrates do not inhibit caspase-3 unlike other caspase substrates or FLICA assays permitting multiday analysis. Consisting of a high-affinity DNA dye linked to a DEVD caspase-3 substrate moiety, the cell permeable non-fluorescent substrate is cleaved by caspase-3 in apoptotic cells to emit fluorescence, allowing detection of both caspase-3 activity and apopototic nuclear morphology. Suitable for flow cytometry, fluorescence microscopy, or live cell imaging systems, staining is compatible with fixation and permeabilization for immunostaining. Biotium Coelenterazine E
Coelenterazine e is a synthetic derivative of coelenterazine. It is a superior substrate for Renilla luciferase generating nearly 40% more light, as well as has an initial light intensity more than 7 times higher than native coelenterazine. Coelenterazine e/Renilla luciferase has two emission peaks centered at 418 and 475 nm, respectively. Coelenterazine e is also a good substrate for apoaequorin compared with native coelenterazine. The e-aequorin compelx has a faster luminescence rise time and shows two emission peaks at 405 and 465 nm respectively, with the ratio of the peak heights dependent on calcium concentration in the range pCa 5-7.
