Dronedarone Hydrochloride is a non-iodinated amiodarone derivative acting as a multichannel blocker for atrial fibrillation. It inhibits Na+, K+, and Ca2+ currents and is a potent inhibitor of acetylcholine-activated K+ current, rapid delayed rectifier, and L-type calcium current.
- Inhibits human Kv1.5 channel with an IC50 of 2.37 μM.
- Blocks IKr (IC50=3 μM) and ICa-L (IC50=0.18 μM).
- Exhibits state-dependent inhibition of fast Na+ channel current.
- Reduces incidence of ventricular fibrillation (VF) in rats.
- Inhibits carotid artery thrombus formation in vivo.
- Impairs thrombin- and collagen-induced platelet aggregation.
- Reduces expression of plasminogen activator inhibitor-1 (PAI1).
