Salt Sabouraud Dulcitol (SSD) Broth is a selective and non-differential medium used in qualitative screening procedures for the cultivation and selection of Candida auris, an emerging pathogen with multidrug resistance. SSD Broth is used as an enrichment step to support growth Candida auris, which can be present in complex skin samples. Multiple published methods are available and include use of a pre-enrichment step (utilizing SSD Broth) with subsequent subculture onto Salt Sabouraud Dulcitol Agar or a chromogenic medium such as Chameleon™ COLOREX™ Candida Plus. Lauryl Sulfate Broth (LSB) is a selective broth utilized for the qualitative detection of coliforms in samples of sanitary importance. The coliform group of bacteria includes aerobic and facultative anaerobic, Gram-negative, non-spore forming bacilli that ferment lactose and form acid and gas at 35°C within 48 hours. Members of Enterobacteriaceae comprise the majority of this group, but organisms such as Aeromonas spp. may also be included. Mannitol Salt Agar (MSA), is a selective and differential medium used for the isolation, enumeration, and differentiation of staphylococci. Chapman formulated Mannitol Salt Agar to isolate staphylococci by inhibiting growth of most other bacteria with a high salt concentration. Pathogenic strains of staphylococci (coagulase-positive staphylococci) grow luxuriantly and produce yellow colonies with yellow zones. Non-pathogenic staphylococci produce small red colonies with no color change to the surrounding medium. Hektoen Enteric (HEK) Agar is a selective and differential medium used for the isolation of enteric pathogens. Compared to other enteric differentiating media commonly used, HEK Agar increased the isolation rate of Salmonella spp. and Shigella spp. This was accomplished by increasing the carbohydrate and peptone content of the medium in order to counteract the inhibitory effects of bile salts and indicators. Rappaport Vassiliadis Salmonella Enrichment Broth is an enrichment broth utilized for the selective isolation of Salmonella spp. from non-sterile samples. RVSE Broth is used as a selective enrichment broth followed by subculture performed onto Xylose Lysine Deoxycholate (XLD) agar. Malachite green and magnesium chloride act as selective agents, combined with high osmotic pressure and low pH to effectively inhibit non-target competitive organisms. Soya peptone provides a source of nitrogen and potassium phosphate acts as a buffer. Bacteroides Bile Esculin (BBE) Agar is an enriched, selective and differential medium recommended for the isolation and presumptive identification of gram-negative obligate anaerobes, such as Bacteroides fragilis and Bilophila species. Selective inhibition of facultative anaerobes and most gram-negative anaerobic organisms is obtained by the presence of gentamicin and oxgall. Differentiation of the B. fragilis is based on esculin hydrolysis, which produces esculetin and dextrose. Sorbitol MacConkey (sMAC) Agar, also known as MacConkey Agar with Sorbitol, is a selective and differential medium used for the isolation of sorbitol-negative Escherichia coli. March and Ratnam introduced sMAC in 1986 for the isolation of verotoxin-producing (VTEC) Escherichia coli O157:H7 from clinical specimens. VTEC does not ferment sorbitol (i.e. - sorbitol-negative), while non-VTEC Escherichia coli are sorbitol-positive.
