Biotium Tyramide Amplification Kit with HRP Streptavidin and CF640R Tyramide
Tyramide amplification, sometimes called Catalyzed Reporter Deposition (CARD), is an enzyme-mediated detection method that uses the catalytic activity of horseradish peroxidase (HRP) to generate high density labeling of a target protein or nucleic acid sequence in situ. The tyramide amplification method has been reported to increase the sensitivity by up to 100-fold compared to the conventional avidin-biotinylated complex procedure. The kit contains HRP streptavidin CF640R dye tyramide. The dye has an excitation/emission of 642/662 nm. The kit contains sufficient reagent for 50-150 slides.

Recognizes an integral membrane glycoprotein of 150 kDa, identified as CD13 (also known as aminopeptidase-N). The antibody recognizes an extracellular epitope. The CD13 antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of AML, CML in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. CD13 is absent from normal lymphocytes, platelets and erythrocytes. HIF1 (hypoxia-inducible factor 1), a heterodimeric transcription factor complex central to cellular response to hypoxia, consists of two subunits (HIF-1 alpha and HIF-1 beta) which are basic helix-loop-helix proteins of the PAS (Per, ARNT, Sim) family. Fluorescein dioctadecanoate is a non-fluorescent hydrophobic fluorescein derivative that can pass through the cell membrane whereupon intracellular esterases hydrolyze the dioctadecanoate group producing the highly fluorescent product fluorescein. The fluorescein molecules accumulate in cells. Fluorescein dioctadecanoate is a fluorogenic substrate for C18 esterase that generates green fluorescence upon esterase cleavage. This antibody recognizes both the free and protein-conjugated (either soluble or cell bound) form of biotin. This MAb is highly specific to biotin and shows no cross-reaction with other structurally related compounds. It has a very high affinity for biotin and is excellent for use in various amplification techniques. In some applications, localization of biotinylated probes with avidin produces unacceptably high background staining. This antibody recognizes a ~90-95 kDa protein which is identified as cell surface transferrin receptor (CD71), a disulfide-bonded homodimeric glycoprotein of 180-190 kDa (Workshop IV).
