Cell Biolabs’ Total Cholesterol Assay Kit is a simple fluorometric assay that measures the amount of total cholesterol present in plasma, serum, tissue homogenates, or cell lysates in a 96-well microtiter plate format. The assay will detect total cholesterol (cholesteryl esters plus free cholesterol) in the presence of cholesterol esterase or only free cholesterol in the absence of the esterase enzyme. Each kit provides sufficient reagents to perform up to 192 assays, including blanks, cholesterol standards and unknown samples. Sample cholesterol concentrations are determined by comparison with a known cholesterol standard. Cholesteryl esters can be quantified by subtracting the free cholesterol values from the total cholesterol value.

The CytoSelect™ LDH Cytotoxicity Assay Kit provides a colorimetric format for measuring cell cytotoxicity. Cells can be plated and then treated with compounds or agents that affect cell viability. Upon cell death, lactate dehydrogenase (LDH), a soluble enzyme found in the cytoplasm, is released into the growth media. In the presence of lactate substrate, LDH converts lactate to pyruvate and generates nicotinamide adenine dinucleotide (NADH). The WST-1 molecule is converted to the orange formazan form. An increase in cell cytotoxicity is accompanied by increased LDH release and increased colorimetric signal. The assay principles are basic and can be applied to most eukaryotic cell lines, including adherent and non-adherent cells and certain tissues, depending on LDH expression levels. The LDH Cytotoxicity Reagent can be used to detect cytotoxicity in mammalian cells. The kit contains sufficient reagents for the evaluation of 960 assays in 96-well plates.

Chemotaxis, a type of cell migration, is a highly integrated, multistep process that orchestrates embryonic morphogenesis, tissue repair and regeneration. The initial response of a cell to a migration-promoting agent is to polarize and extend protrusions in the direction of the attractant; these protrusions can consist of large, broad lamellipodia or spike-like filopodia that are driven by actin polymerization and can be stabilized by extracellular matrix (ECM) adhesion or cell-cell interactions. The CytoSelect™ Cell Migration Assay Kit utilizes polycarbonate membrane inserts (8 µm pore size) to assay the migratory properties of cells. The kit contains twelve 3 um polycarbonate membrane inserts in a 24-well plate, through which migratory cells are able to pass. Cells are quantified using the provided fluorometric dye. The 3 µm pore size is optimal for measuring the migration of leukocytes.

The Oxygen Radical Antioxidant Capacity (ORAC) Assay is a classic tool for measuring the antioxidant capacity of biomolecules from a variety of samples. The ORAC Activity Assay is based on the oxidation of a fluorescent probe by peroxyl radicals by way of a hydrogen atom transfer (HAT) process. Peroxyl radicals are produced by a free radical initiator, which quenches the fluorescent probe over time. Antioxidants present in the assay work to block the peroxyl radical oxidation of the fluorescent probe until the antioxidant activity in the sample is depleted. The remaining peroxyl radicals destroy the fluorescence of the fluorescent probe. The OxiSelect™ In Vitro ROS/RNS Assay Kit is an assay for measuring the total free radical presence of a sample. The assay employs a proprietary quenched fluorogenic probe, DCFH-DiOxyQ), which is based on similar chemistry to the popular DCFH-DA. The DCFH-DiOxyQ probe is first primed with a quench removal reagent, and subsequently stabilized in the highly reactive DCFH form. In this reactive state, ROS and RNS species can react with DCFH, which is rapidly oxidized to the highly fluorescent 2’, 7’-dichlorodihydrofluorescein (DCF) . The DCFH-DiOxyQ probe can react with hydrogen peroxide, peroxyl radical, nitric oxide, and peroxynitrite anion. These free radical molecules are representative of both ROS and RNS, thus allowing for measurement of the total free radical population within a sample. The kit has a detection sensitivity limit of 40 nM for H2O2. Each kit provides sufficient reagents to perform up to 5x96 assays, including standard curve and unknown samples.
