Cell Biolabs Inc CytoSelect™ WST-1 Cell Proliferation Assay Reagent, 960 assays
The CytoSelect™ WST-1 Cell Proliferation Assay Reagent provides a colorimetric format for measuring and monitoring cell proliferation. Cells can be plated and then treated with compounds or agents that affect proliferation. Cells are then detected with the proliferation reagent, which is converted in live cells from WST-1 to the formazan form in the presence of cellular NADH and an electron mediator. An increase in cell proliferation is accompanied by increased signal, while a decrease in cell proliferation (and signal) can indicate the toxic effects of compounds or suboptimal culture conditions. The assay principles are basic and can be applied to most eukaryotic cell lines, including adherent and non-adherent cells and certain tissues. This reagent can be used to detect proliferation in bacteria, yeast, fungi, protozoa as well as cultured mammalian and piscine cells. The reagent is sufficient for the evaluation of 960 assays in 96-well plates or 192 assays in 24-well plates.
Biotium Resazurin, sodium salt
Resazurin is the N-oxide of the fluorescent dye resorufin and used to detect reductive activities in cells. It is widely used for measuring cell proliferation and mitochondrial metabolic activity. Resazurin itself is non-fluorescent until reduced to the highly red fluorescent resorufin (Ex/Em: 563/587 nm) in the presence of reductants like NADPH or NADH and the enzyme diaphorase. Resazurin can therefore be used to detect NADH, NADPH, or diaphorase levels. Furthermore, the resazurin /diaphorase /NADPH system can be used to detect any biochemical or enzyme activity involved in generating NADH or NADPH. Although resazurin is available from several suppliers, trace amounts of the resorufin contaminant often make the material unsuitable for bioassay applications. This high-grade resazurin has a minimal background fluorescence. Biotium JC-1 (IODIDE SALT)
JC-1 (5,5',6,6'-Tetrachloro-1,1',3,3'-tetraethylbenzimidazolylcarbocyanine, iodide) is a mitochondrial dye that stains mitochondria in living cells in a membrane potential-dependent fashion. JC-1monomer is in equilibrium with so-called J-aggregates, which are favored at higher dye concentration or higher mitochondrial membrane potential. The monomer JC-1 has green fluorescence ( Em = 527 nm), while the J-aggregates have red fluorescence ( Em = 590 nm). Therefore, it is possible to use fluorescence ratioing technique to study mitochondrial membrane potentials. JC-1 is particularly useful for apoptosis studies.
