Biotium Resazurin, sodium salt
Resazurin is the N-oxide of the fluorescent dye resorufin and used to detect reductive activities in cells. It is widely used for measuring cell proliferation and mitochondrial metabolic activity. Resazurin itself is non-fluorescent until reduced to the highly red fluorescent resorufin (Ex/Em: 563/587 nm) in the presence of reductants like NADPH or NADH and the enzyme diaphorase. Resazurin can therefore be used to detect NADH, NADPH, or diaphorase levels. Furthermore, the resazurin /diaphorase /NADPH system can be used to detect any biochemical or enzyme activity involved in generating NADH or NADPH. Although resazurin is available from several suppliers, trace amounts of the resorufin contaminant often make the material unsuitable for bioassay applications. This high-grade resazurin has a minimal background fluorescence. Fura-2, AM ester is an intracellular fluorescent calcium indicator, and the preferred dye for ratiometric imaging microscopy. The membrane-permeant AM ester can be loaded into cells via incubation, but due to limited water solubility, a mild detergent Pluronic F-127 is often used to facilitate cell loading. Fura-2, AM ester itself does not bind Ca2, but intracellular esterases readily hydrolyze it to Fura-2. Upon calcium binding, the excitation maximum of Fura-2 undergoes a blue-shift from 363 nm (no Ca2) to 335 nm (high Ca2) while the emission maximum is relatively unchanged. Measure by flow cytometry, fluorescence microscopy, or certain fluoresence microplate readers.
