Activated by phosphatidylinositol 3,4,5-trisphosphate (PIP3), AKT proteins regulate numerous cellular functions and are critical for tumor cell survival, inhibiting apoptosis and regulating the mTOR pathway. AKT1 mutations are associated with cancer types, including breast, colorectal, and ovarian cancers, as well as disorders like Proteus syndrome and Cowden syndrome. Gene amplification or overexpression of this protein has been observed in cancers like prostate, bladder, and breast tumors. Alternate splice variants have been characterized, including one isoform that lacks the intermembrane region and is secreted outside the cell, where it modulates the activity of the membrane-bound form. Other splice variants have been reported but are not fully characterized.
