Can Get Signal is a solution containing an accelerator for antigen-antibody reactions. This reagent improves sensitivity, specificity, and signal-to-noise ration (S/N) for Western blotting, dot blotting, enzyme-linked immunosorbent assay (ELISA), etc. Solutions 1 and 2 refer to the reactions of the primary and secondary antibodies, respectively.

Mouse IgG TrueBlot ULTRA is provided as 1000X solution. We recommend incubating the blots from minigels in sealed bags with minimal volume (2-3 mLs). If used conservatively at 2.5mls/blot will yield enough reagent for 20 blots. Note that there are three key procedural considerations: 1. Protein A or G beads may be used with the mouse, goat and sheep TrueBlot secondaries, but not with the rabbit TrueBlot secondary. Use of protein A or G beads with the rabbit TrueBlot will result in contaminating bands. 2. Immunoprecipitate should be completely reduced. 3. BLOTTO/Milk should be used as the blocking protein for the immunoblot. We recommend using a large bore pipet to pipet up the liquid for use. To achieve best results when detecting mouse IgG1 subtypes, we recommend performing a dot blot or titration to determine the optimal dilution. Specific conditions for each protein and antibody combination should be specifically optimized by the end user.

CellTag 520 Stain is a visible fluorescent, non-specific cell stain that provides accurate normalization to cell number for In-Cell Western™ Assay applications. The stain accumulates in both the nucleus and cytoplasm of permeabilized cells, and provides linear fluorescent signal across a wide range of cell types and cell numbers. CellTag 520 Stain is detected in the 520 nm channel. CellTag 520 Stain is applied to the cells during incubation with an IRDye Secondary Antibodies (e.g., IRDye 680RD or IRDye 800CW), and enables accurate measurement of target protein levels with much higher throughput than Western blotting.
